Полученные фрагменты опухоли пациента имплантировали мышам линии Balb/c Nude в подкожный сайт, получив первую генерацию ксенографта. Исследование ксеногенной модели проводили на четвертой генерации. Ткани PDX-модели и пациентской опухоли анализировали с помощью гистологического, иммуногистохимического и молекулярно-генетического методов. Для иммуногистохимического исследования использовались следующие антитела: ER (рецептор эстрогена); PR (рецептор прогестерона); HER2/ neu (рецептор человеческого эпидермального фактора роста 2-го типа); Ki-67 (маркер пролиферации). Молекулярно-генетическое исследование генов BRCA1 и BRCA2 выполняли методом полимеразной цепной реакции в режиме реального времени.</p></sec><sec><title>Результаты</title><p>Результаты: Проведенное гистологическое исследование выявило соответствие гистотипа опухолевой ткани пациентки Е. и ксенографта. Иммуногистохимический анализ данных образцов опухолей показал отсутствие экспрессии маркеров ER, PR и HER2. При этом в опухолевом материале пациентки было выявлено 80% клеток, экспрессирующих Ki-67, а в ксеногенном материале – 90%. Также в исследуемом опухолевом фрагменте пациентки и соответствующем ему ксенографте не было обнаружено мутаций в генах BRCA1 и BRCA2.</p></sec><sec><title>Заключение</title><p>Заключение: В ходе данной работы была проведена оценка клинически значимых маркеров PDX-модели ТНРМЖ, которая показала совпадение с соответствующими характеристикам опухоли пациентки Е. Таким образом, нами получена PDX-модель, которую можно применять как для фундаментальных, так и доклинических исследований в области онкологии, в том числе для изучения противоопухолевой эффективности новых препаратов в отношении ТНРМЖ, а также для выявления механизмов чувствительности или резистентности к различным терапевтическим воздействиям.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Objective</title><p>Objective: To evaluate a correlation of clinically relevant markers between a patient-derived xenograft (PDX) model and triple-negative breast cancer (TNBC).</p></sec><sec><title>Materials and methods</title><p>Materials and methods: For the PDX model of TNBC, we used tumor tissue from patient E. who presented with right-sided breast cancer at the National Medical Research Centre for Oncology (Rostov-on-Don, Russian Federation). The tumor fragments were subcutaneously implanted into BALB/c nude mice; thus, the first-generation xenograft was obtained. The xenogeneic model was studied on the fourth generation. The PDX model tissues and the patient’s tumor were analyzed by histological, immunohistochemical, and molecular genetic methods. For the immunohistochemical study we used antibodies against ER (estrogen receptor), PR (progesterone receptor), HER2/neu (human epidermal growth factor receptor 2), and Ki-67 (proliferation marker). Molecular genetic testing of the BRCA1 and BRCA2 genes was performed using real-time polymerase chain reaction.</p></sec><sec><title>Results</title><p>Results: The histological examination demonstrated a correlation between the histotype of the original tumor tissue and the xenograft. Immunohistochemical analysis of the tumor samples revealed no expression of ER, PR, and HER2 markers. We identified 80% of Ki-67-expressing cells in the patient’s tumor and 90% in the xenograft. Furthermore, no mutations in the BRCA1 and BRCA2 genes were found in the patient’s tumor and the corresponding xenograft.</p></sec><sec><title>Conclusions</title><p>Conclusions: We evaluated clinically relevant markers in the PDX model of TNBC and found the correlation with the corresponding characteristics of the patient’s tumor.
